Sequence variation of bovine mitochondrial ND-5 between haplotypes of composite and Hereford Breeds of beef cattle

چکیده

The aims of the study were to: Investigate polymorphisms in the ND-5 region of bovine mitochondrial DNA in the composite and purebred Hereford herds from the Wokalup selection experiment, sequencing and compare the sequences between haplotypes and published sequence from Genebank. A total of 194 Hereford and 235 composite breed cattle from Wokalup Research Station were used in this study. The mitochondrial DNA was extracted using Wizard genomic DNA purification system from Promega. ND-5 fragment of mitochondrial DNA was amplified using PCR and continued with RFLP. Each haplotypes were sequenced. PCR products of each haplotype were cloned into pCR II, transformed, colonies selection, plasmid DNA extraction continued with cycle sequencing. Polymorphisms were found in both breeds of cattle in ND-5 region of mitochondrial DNA by PCR-RFLP analysis. Sequencing analysis confirmed the RFLPs data. © 2002 Jurusan Biologi FMIPA UNS Surakarta

Mitochondrial DNA polymorphisms and fertility in beef cattle.

Four hundred and twenty-two beef cattle of two different breeds (purebred Hereford and composite multibreed) were characterized by polymerase chain reaction-restriction fragment length polymorphism, using the restriction enzymes ApaI, AvaII, HindIII, PstI, SpeI, SspI and TaqI in two regions (the D-loop and the ND-5 gene) of mitochondrial DNA. The association between molecular haplotypes and rec...

Sequence Variations of Mitochondrial DNA Displacement-Loop in Iranian Indigenous Sheep Breeds

Mitochondrial DNA (mtDNA) has been used extensively to study population genetics because it has the unique features of maternal inheritance, a relatively fast rate of evolution and lack of recombination. A total of 82 unrelated sheep from 10 Iranian indigenous sheep breeds were investigated to determinate the maternal genetic diversity using a sequence of a 685 bp segment of the displacement lo...

Comparison of growth traits of eight beef cattle breeds in the Czech Republic [in German]

Eight beef cattle breeds, Angus (A), Blonde d' Aquitaine (BA), Charolais (C), Czech Pied (CP), Hereford (H), Limousin (L), Piemontese (P) and Simmental (S), were analysed for the following calf traits: birth weight (BW), 210and 365-day weight ( 210W, 365W) and average daily gains from birth to 210 days (ADG1), from 210 to 365 days (ADG2) and from birth to 365 days (ADG3). CharaCteristiCs of phenogroups of the a blood group system in Cattle of beef breeds**

phenotypes of four beef breeds (Charolais, hereford, limousin and piemontese) were determined based on the transmission of antigens from parents to offspring in the a blood group system of cattle. the effective number of alleles per locus (e) and the degree of heterozygosity (h) were calculated for different breeds based on the frequency of these phenogroups. most a-phenogrups (18) were found i...
